How to do Panasonic 42" LCD L42U30D calibration...??

Recently i have purchased the Panasonic L42U30D model LCD..... Kindly suggest the calibration settings for the LCD.
I am not able to run .mkv files on this LCD.. which is the best convertor to convert the mkv files and to which format??

Kindly help...
